High-Density Vero Cell Perfusion Culture in Single-Use Vessels
Vero cells are anchorage-dependent cells that are widely used as a platform for viral vaccine production. In stirred-tank bioreactors, they are ordinarily grown on microcarriers. However, Fibra-Cel® disks are a promising alternative attachment matrix with a high surface-to-volume ratio. They provide a three-dimensional environment that protects cells from damaging shear forces, helping to achieve high cell densities.  We will discuss cultivating Vero cells in single-use vessels pre-packed with Fibra-Cel using the BioFlo® 320 bioprocess control station in perfusion mode ensuring a consistent supply of nutrients and the removal of toxic byproducts, and how a cell density of approximately 43 million cells per mL was achieved.

Redox Potential Monitoring for Improved Anaerobic Fermentation Using Fermentation Single-Use Vessels
In fermentation, redox potential is an important physiochemical factor that measures the tendency of the culture medium to acquire electrons. It can directly influence bioprocess efficiency. For example, in the anaerobic fermentation of Clostridium to produce industrial solvents, the redox potential relates to the intracellular ratio of NAD(P)+ to NAD(P)H, which in turn influences the production of solvents, including butanol.  We will discuss how C. beijerinckii was cultivated under anaerobic conditions in fermentation single-use vessels, and how redox potential of the fermentation broth was monitored online using ISM® redox sensors and controlled by adding sodium sulfide.  Bacterial growth and butanol production drastically increased compared to a process without redox potential control.

Producing a successful bioprocess product requires many phases of development and validation. During each phase, from expression construct or virus engineering all the way to manufacturing, it is critical to keep careful track of archived samples, data, and protocols for future recall/audit.
